Types of membrane preparations used

  1. Crude homogenates (2)
  2. Plasma membrane preparations (P2 fraction, 3)
  3. Sucrose density gradient enriched receptors (1)
  4. Commercially available membranes, which includes:
    • Receptors cloned into mammalian cells (Perkin Elmer, Euroscreen, Cerep)
    • Receptors cloned into Sƒ9 insect cells co-expressing mammalian G-proteins

Types of Assay buffers

  1. Lazareno and Birdsall buffer (2) 20 mM HEPES, 100 mM NaCl, 5 mM MgCl2, pH 7.4.
  2. Buffers with HEPES replaced by 50 mM Tris HCl (1,9)
